It is Summer 1938. Three years after the events of The Ark of the Covenant.
Archaeology professor Dr. Henry "Indiana" Jones, Jr. (Harrison Ford) and museum curator Dr. Marcus Brody (Denholm Elliott) are invited by Walter Donovan (Julian Glover) to his mansion. Donovan is a generous donator to the museum. He has discovered fragment of an ancient stone marker dating to the 12th century. Indy reads the marker, and to his surprise it details "the cup that holds the blood of Jesus Christ" - the Holy Grail, whose can grant immortality.
Donovan's project team is now searching for the second marker, left behind by a knight who had found the Grail, somewhere in Venice. Unfortunately the project leader and all his research have vanished. To Indy's surprise, the missing project leader is his father Professor Henry Jones, Sr. (Sean Connery). Indy suddenly remembers the brown paper package from Venice – his dad's diary, which contains map and information about the Holy Grail.
Indy and Marcus fly to Venice to meet Dr. Elsa Schneider (Alison Doody), his father assistant. They go to where Henry Sr. disappeared, the Venice library, which was once a church in ancient Roman times. Indy discovers that the library is the tomb of the knight. Indy digs the floor and finds a tunnel. Exploring the tunnel with Elsa, Indy finds an ancient casket and a knight's shield, the second marker. Indy learns that his father is imprisoned in a castle in Germany.
Indy and Elsa fly to the German castle and rescue Henry Sr., but Indy is betrayed by Elsa and Walter Donovan. Indy and his father are imprisoned in the castle but they manage to break out and escape from German soldiers. They go back to Berlin, where Donovan and Elsa take the diary for Hitler. Adolf Hitler and the Nazis plan to use the Holy Grail for global world domination for their super-race. Indy treks to Berlin, finds Elsa, and takes the diary back.
